Dulaglutide

Overview

Dulaglutide (Trulicity) is a long-acting GLP-1 receptor agonist engineered by Eli Lilly as a fusion protein. Its first FDA clearance, in September 2014, covered glycemic control in type 2 diabetes; a second, in February 2020, addressed cardiovascular risk, specifically the reduction of major adverse cardiovascular events where type 2 diabetes coincides with established cardiovascular disease or a cluster of risk factors. Architecturally it comprises two identical disulfide-linked chains; within each chain a DPP-4-resistant human GLP-1 analog fragment is covalently joined through a synthetic peptide linker to a human IgG4 Fc fragment, a layout that lengthens half-life and supports once-weekly administration. Lyochem supplies Dulaglutide as a lyophilized reference standard, characterized as a folded ~59.7 kDa protein rather than a linear peptide. Identity and purity are established by SEC and RP-HPLC for aggregate and main-species content, reduced/non-reduced SDS-PAGE to confirm the disulfide-linked homodimer and chain mass, and ESI/LC-MS mass characterization; residual water content is documented per lot so the certified quantity reflects net protein.
