Overview
Humanin (HN) is a 24-residue peptide of the mitochondrial-derived-peptide (MDP) class, sequence MAPRGFSCLLLLTSEIDLPVKRRA, encoded by a short open reading frame within the mitochondrial 16S rRNA gene (MT-RNR2) rather than by nuclear DNA. It was the first MDP to be described, reported by Hashimoto and colleagues in 2001 (PNAS 98:6336) from a screen of an Alzheimer's-patient occipital-lobe cDNA library for factors antagonizing neuronal apoptosis triggered by the mutant APP variant V642I. That founding discovery established the mitochondrial genome as a source of bioactive signaling peptides. The lone Cys can oxidize, which is why identity rests on ESI-MS against the 2687.28 Da target plus LC-MS/MS mapping of the MAPRGFSCLLLLTSEIDLPVKRRA sequence: the mapping localizes the residue and any mass shift at that position, so an oxidized or adducted form is distinguishable from the intended reduced peptide rather than passing on retention time alone.
Hold the sealed lyophilized peptide at -20 C, desiccated and light-protected. Reconstitute just before use, and because a cysteine-containing peptide is prone to oxidation in solution, avoid extended aqueous storage and repeated freeze-thaw that would erode the RP-HPLC purity documented on the COA.
